The primary Modifier key you would use for this is "Control". Make sure your Global Setting is set to Off and fill out the desired modifier keys as desired. If you, for some reason, use the standard point-and-click style, this also works. This works the exact same for players who use On Release as their standard and wish to use Quickcast as their Modifier combination, just use the Hero Abilities (Quick Cast) menu instead, and set your Global Setting to On Release. I click CTRL + W and it'll give me the indicator of where my cast will land. However, sometimes I will be clearing waves and want a precise Blizzard to cover all the creeps. My regular method of casting spells is set to Quick Cast, so if I am playing Jaina and my tank stuns a noob, I can faceroll WEQ and cast everything in an instant.

When I save these settings and get into a game, I now have the option of both Quick Cast as well as Quick Cast on Release through simple hotkeys without changing anything.

I can use Control + W (Control being my modifier key) to use the On Release setting instead, allowing me to essentially use two hotkey profiles with their unique advantages with the simple use of a modifier key. However, sometimes I require precision, for example when using Piercing Shard Ice Lances on a Siege Camp, or Blizzard on a Minion Wave. My global setting is "On", meaning my abilities will cast as I click the hotkey. Illustrative Example: When I play Jaina, I want to be able to cast my spells as fast as possible to burst as hard as possible. You can easily set this up in the Hotkey menu following this little guide. This allows you to have your prefered ability activation method, but gives you access to a secondary method on-demand. TLDR: It is possible to set up a modifier key to switch between Hotkey settings (On Release, On, or Off).
